Effects of 5-Fluorouracil on the Parotid Glands of Mice An Electron Microscope Study



Abstract
A total of one hundred and forty male mice weighing 20gm were used throughout this experiment.
Experimental mice were given an intramuscular injection of 5-fluorouracil 50mg per kg of body weight.
Animals were sacrificed 1, 3, 5,7, 10 and 14 days after injection of the 5-fluorouracil.
Since body weight and side action of the mice have shown that 5-fluorouracil effect "was most pronounced on day 5 after the last injection of 5-fluorouracil, electron microscope study was done using only those mice on day 5 of experimentation.
For electron microscopy, the animals were perfused with 2% paraformaldehyde solution in 0.1M phosphate buffer under anesthesia with 3.5% chlorohydrate.
The fixative was prewarmed to 36.5¡ÆC and perfusion made by an intracardiac approach using a 50m1 syringe filled with the fixative, In general approximately 100 to 150ml of the fixative was perfused in five minutes.
Pieces of the parotid gland were taken out and fixed further in a mixture of 1 % paraformaldehyde and 1 % osmium tetroxide in the same buffer. Following washing and dehydration in graded ethanols, the tissues were embedded in a mixture of epoxy resin in a routine manner.
Ultrathin sections were made on a LKB 8801 ultramicrotome, stained with 2% uranyl acetate and lead citrate.
Observations were made in a Hitachi Hu-Ile and Hitachi HS-8 electron microscope.
Electron microscope appearance of acinar cells from the 5-fluorouracil treated animals showed following changes.
1. In general, the cells appeared to be smaller, containing fewer secretory granules and less organized cytoplasmic organelles.
2. Though in most cells the basal rough endoplasmic reticulum was well-developed, there were occasional cells in which the"rough endoplasmic reticulum appeared less tightly organized. Elements of the rough endoplasmic reticulum tended to become confluent, producing.a labyrinthino appearance of the organelle.
3. Mitochondria of cell tended to be smaller, more rounded in appearance; and contained a less. electron dense matrix and fewer cristae:
4. The Golgi complex of acinar cells from 5-fluororacil treated animals showed in general a marked reduction in its degree of organization. This included a shortening of the lamellar elements which were composed of only a few stacks.
5. The number of transport vesicles also appeared reduced in comparison to that of control cells. 6. Lipid granules were occasionally-, fqund in the normal animals, but they were definitely increased in number at the base of acinar cells from 5-fluorouracil treated mice.
7. The structure of nucleoli was difficult to judge, as many of them were-large-and -presented-a normal appearance. However, there were occasional nucleoli which were very small, being comprised of a rather simple aggregates. of nucleolonema, and associated chromatin masses. Such a nucleoli resembled those that bad been seen in other antimetabolite treated cells.
